DNA shearing

최적의 NGS 라이브러리 준비에서 가장 중요한 요소 중 하나는 시퀀싱 할 DNA의 품질입니다. 신뢰할 수 있는 NGS 결과를 얻으려면 먼저 DNA가 적절한 사이즈로(sequencing platform에 따라 다름)로 일관성있게 shearing되어야 합니다. Bioruptor는 NGS workflow에 필수적인 우수한 수율, 크기 및 일관성을 제공합니다.

Protein extraction

생화학 실험에서는 조직이나 포유류, 효모 및 박테리아 세포에서 단백질을 추출해야하는 경우가 많습니다. 고품질의 단백질을 높은 수율로 얻는 것은 PAGE, Western blot, 질량 분석 또는 단백질 정제와 같은 downstream protein characterization에 중요합니다. Bioruptor를 이용하면, 초음파 처리 기반으로 세포를 파쇄하게 됩니다. 이 방식은 한 번에 조직 및 배양 세포를 효율적으로 파쇄하고 균질화할 수 있어 식별 가능한 펩티드 및 단백질 수를 크게 향상시킵니다. 즉, 고품질 단백질 제공으로 proteome 전체의 발견을 돕습니다.

Protein seeding and aggregations

Amyloid-like aggregates (amyloid fibrils)의 형성은 정상적인 proteome의 기능을 방해하고 신경 퇴행성 질환의 독성에 기여합니다. Amyloid 형성 및 세포 증식 특성 연구에서, 초음파 처리는 새로운 응집체 성장을 위한 아밀로이드 섬유 분해에 쓰입니다. 이 때 아밀로이드 섬유가 분해되면서 응집체 형성이 촉진 되면서, seed가 생성되게 됩니다. Bioruptor는 초음파 처리 및 온도 처리를 통해 seeding material을 표준화하고 생성하는 데 많은 이점이 있습니다.

Liposome preparation

초음파 처리는 liposome preparation에 사용되는 가장 일반적인 방법 중 하나입니다. Bioruptor를 사용하면 직접적인 접촉이 필요치 않아 시료 오염을 방지할 수 있습니다. 또한 정확한 온도 제어를 통해 과열 또는 산화로 인한 손상으로부터 지질을 보존할 수 있습니다.
